English Abstract

The present study was conducted to investigate the active constituents found in ethanolic extract of Sesamum indicum defatted seeds, and studies it effect on the serum levels of lipid and cytogenetic changes in bone marrow cells in laboratory mice (in vivo).

The cytotoxic effects of sesame extract on three types of cancer cell lines and one normal embryo rat cell line was also investigated.

Results of general biochemical detection showed that ethanolic extract of sesame seeds contain phenols, tannins, saponines, glycosides, alkaloids, coumarines, and flavonoids, while terpines and steroids were absent.

The qualitative and quantitative determination of sesamin bioactive compound using High Performance Liquid Chromatography (HPLC) analysis was carried out and compared with standard sesamin.

Results showed that the concentration of sesamin in the ethanolic extract was 79.9% of ethanolic extract according to total peak area.

The effect of ethanolic extract of sesame seeds on the serum lipid profile [total cholesterol, triglycerides, High Density Lipoprotein (HDL), and Low Density Lipoprotein (LDL)] were evaluated in laboratory mice (in vivo) when fed with high and low fat diet.

Sesame extract at concentration 500 mg/kg body weight (B.W) showed significant reduction (p≤0.05) in the level of total cholesterol (56.14 mg/dl), triglycerides (61.12 mg/dl), and low density lipoprotein LDL (29.97 mg/dl) and significant increase in the level of high density lipoprotein HDL (18.00 mg/dl) in comparison with high fat group which recorded 80.34 mg/dl, 77.50 mg/dl, 48.09 mg/dl, and 16.80 mg/dl respectively.

Significant increase was recorded in mitotic index (MI) at concentrations 500 mg/kg(B.W) in mice fed with high fat diet (20.17%), while in mice fed with low fat diet, extract showed significant increase in MI at both concentrations 250 and 500 mg/kg (B.W) (9.42% and 13.47%) respectively.

Summary Sesame seeds extract at both concentrations 250 and 500 mg/kg (B.W) showed significant increase in the level of deletion and ring chromosomes at both high and low fat diet treatment.

Results of in vitro growth inhibition (GI) of sesame extract against Human larynx epidermoid carcinoma (Hep-2), Ahmed- Mohammed- Nahi-2003 (AMN-3), Rhabdomyosarcoma (RD), and normal embryo rat (Ref) cell lines demonstrated that the growth of these cells significantly decreased when compared with control cultures (untreated), and the effects were dose, as well as, time- dependent for each Hep-2, AMN-3, and Ref cells.

A clear cytotoxic activity was observed during an exposure time of 72 hrs at a concentration 1000 μg/ml which reached (85.83, 40.06, and 20.20) respectively, while the percentage of inhibition during exposure time of 48 hrs recorded 31.03 at a concentration of 1000μg/ml for RD cells.
