English Abstract

The fungal isolates Pleurotus ostreatus and Pleurotus sapidus were evaluated for extracellular laccase production.

The two isolates were subjected to screening process to select the highest extracellular laccase production.

Results indicated that P.

ostreatus was showed the highest productivity of the enzyme.

Optimization studies were done by the solid state fermentation and revealed that the barley bran (7.5gm) was the best substrate, at initial moisture ratio 1:1.5 (w/v), using mineral salts solution as moisturizing solution with inoculums size 3 5 mm agar disk plugs at 25°C for 10 days to yield 2.93 U/mg for Laccase enzyme specific activity.

Extracellular laccase produced from Pleurotus ostreatus was purified by two steps including dialysis and DEAE- cellulose ion exchange chromatography.

The overall purification folds were 8.29 times with a 36.8% recovery.

The molecular weight of laccase was approximately 62.97 KDa as determined by SDS- PAGE.

The characterization of purified enzyme that the optimum pH for activity was 6.5 and (6.5-7.5) for stability.The maximum activity was observed at 45Cºand the enzyme showed 100% of its activity after 1h when incubated at temperature ranged (15-45) Cº, then the activity decreased with increasing temperature at 85Cº and the enzyme retained 4.41% of the initial activity, results also showed that enzyme retained 60% of its activity after 24h when incubated at optimum pH and temperature.

Laccase activity became 20% after 48 h of incubation.

The ability of P.

ostreatus culture to decolorize textile dyes revealed that the fungus grew well and completely decolorized Blue | P a g e 8 dye H3R with concentrations (50, 100, 150 and 200) and approximately 98% with 250 ppm concentration in comparison with yellow and Red dyes were very slowly processed, dye decolorization was appeared only at 50 ppm concentration.

The ability of partial purified laccase to decolorize of textile dyes showed that the blue H3R dye was completely decolorized in all concentrations within first min while yellow FG and red 3B dyes were decolorized in different percentage.
