English Abstract

Eclipta alba plants were collected from Tigris River banks for the period from November 2011 until January 2012.

The percentage of moisture was measured for the plant which was reached to 50%.

Different solvents were used methanol 70% , absolute ethanol , hexane 98% and hot and cold water in the process of extraction of raw materials which were (76, 55, 53, 11, 5)%, respectively. The qualitative chemical detection were done for the active compounds in the crude extract, the tests showed a positive result for the detection of the comarines, phenolic, tannins , Flavonoid, glycosides , alkaloids, resins, and volatile oils .The comarines were studied in the crude extract.

The pH for methanol extract was measured which was 6. The wedelolactone was selected as an active material and the silica gel was used in the partial purification of this material, the results of the absorbance measurements, showed there was one major peak occurred between fractions (7-22).

Then these fractions were collected and concentrated for further qualitative tests to certify the degree of the wedelolactone purity.

The detection measurements were carried for melting point, which reached 328 °C, and its ability to dissoluble in solvents as (Ethanol, Methanol, Hexane, Chloroform and Benzene) was high, while it was less soluble in water and Diethyl ether, and difficult to dissolve in acetone, but it can be soluble in the acetone when mixed with Toluene and Formic acid. The thin layer chromatography showed positive results throughout emerge, the purple color under ultraviolet light, and the Rf value were measured, which reached 0.56 cm, The degree of absorbance was identified through the appearance of sharp peak at 351 nm., and the FTIR testing showed the effective groups for the wedelolactone and they are largely compatible with the structural composition of wedelolactone.

HPLC test showed the high compatibility between the sample and the composite of standard one for wedelolactone in its form and the retention time, which was 2.157 for the sample in comparisons with the standard 2.163, the concentration of the wedelolactone was calculated which was 99.27. A study of LD 50 result showed the safety of wedelolactone at wide doses between (500-3000) mg/kg.

And showed different levels of activity against G+, G- bacteria in vitro. while in vivo study of wedelolactone showed heterogeneous results, when used in different ways, when it was compared with control models, it gave the best results when used locally (external use ointment), but its effectiveness as antibacterial (internally use) was not at the required level through its negative impact on the white blood cells numbers (monocyte, lymphocyte and granulocyte). The tissues study showed the necrotic changes effects of wedelolactone on liver, spleen and intestine tissues plus some white blood cells infiltration.
