English Abstract
Crude β-galactosidase extracts of Zingiber officinale and Alliumn sativum were investigated for the activity β-galactosidase.
The enzyme in the extract was characterized in term of pH, temperature and enzyme kinetic.
The crude β-galactosidase extract of Zingiber officinale and Alliumn sativum used also in hydrolysis of lactose in milk and whey.
The enzyme activity was measured by its ability to hydrolyze the substrate 2-nitrophenyl β -D-galactopyranoside (ONPG).
The crude enzyme extract of Zingiber officinale was found to has maximum activity at pH =5.0 and 60°C.
While extract of Alliumn sativum was found to has maximum activity at pH=4.0 and 500C.
Enzyme kinetics in both extracts were indicated to have a potential activities for example, the km and Vmax values of enzyme extract of Zingiber officinale were 2.08 mM and 0.55µmol/min, respectively while 3.2mM and 1.1 µmol/min, respectively in Alliumn sativum.
The application of crude enzyme extract of both plants for lactose hydrolysis in milk showed a significant decreased in the lactose content 30-45% in raw milk, while 50-60% of lactose hydrolyzed in skimmed milk.
The findings indicate the potential use crude enzyme extract of Zingiber officinale and Alliumn sativum for the production of low/delactosed milk for lactose-intolerant population.
However, 55-60% of lactose hydrolyzed in whey.
These results encourage to use their crude enzyme extract in food industry and in manufacturing syrup sweeteners.
