English Abstract

Chitinolytic enzymes are receiving attention due to their role in biocontrol, medicine and agriculture.

A total of 20 strains of bacterial isolates with extracellular chitinolytic activity were isolated from 50 different dead arthropods collected from different regions of AL-Tafilah, South Jordan.

Bacterial isolate A7 was found to produce the chitinase with higher activity and biocontrol property.

Isolate A7 was identified biochemically and morphologically as well as by 16S rRNA gene sequencing as Bacillus atrophaeus.

Cultivation of B.

atrophaeus in chitin liquid medium resulted in production of chitinase with total activity of 68.9 mU/ml and specific activity of 2670 mU/mg protein.

Calculation of enzyme activity was carried out by DNS method.

The enzyme retained its activity at temperature between 20 ºC to 50 ºC with optimum activity at 40 ºC.

The enzyme was most active between pH 4 and pH 9.

It was relatively stable between at neutral pH ranges, beyond these pH ranges, its activity was rapidly decrease or lost.

This study pointed out the importance of microbial chitinase in controlling insect development and metamorphosis.

atrophaeus A7 inhibits development of D.

melanogaster larvae and it pupation with LD50 (24.1 ± 0.9 mU/ml) and delay the development at low concentrations.
