حصل على 75 عزلة تعود إلى بكتريا Pseudomonads aeruginosa من أصل 120 عينة جمعت من حالات مرضية مختلفة.
أظهرت نتائج التحري عن جين الضراوة لانزيم LasB (Pseudolysin) Elastase في بكتيريا P.
aeruginosa إلى وجود هذا الجين في 63 عزلة و بنسبة 84 % و كان الحجم الجزيئي للجين 300 زوج قاعدة بعد ترحيله كهربائيا.
حلل التسلسل التتابعي لجين LasB في بكتريا P.
aeruginosa بينت النتائج أن هنالك طفرات وراثية في دنا الجين LasB، حدثت هذه الطفرات في تسلسل القواعد النتروجينية و كان أغلبها من نوع الاستبدال و الإضافة و الحذف و كانت نسبة التطابق 97 % مع التسلسل التتابعي للجين الأصلي، كذلك تبين من مطابقة التسلسل التتابعي للقواعد النتروجينية المترجمة مع تسلسل الحامض الأميني الذي يشفر عن هذا الجين، لم يكن للطفرات تأثير في نتائج ترجمة البروتين و ذلك لكونها طفرات صامتة.
Abstract EN
Out of 120 isolates from different clinical cases, only 75 were found and confirmed that they belong to the Pseudomonas aeruginosa bacteria.
The result revealed that the LasB virulent gene was present in 63 isolates with 63 % percentage.
The gel electrophoresis showed that the molecular weight of LasB gene was 300 bp.
DNA sequences of LasB gene was done, and the results showed the presence of some gene mutations like substitution, addition and deletion with 97 % identity with the Refseq gene.
From the other side, the results of identities of translated nucleotides sequence with the original sequence of amino acids revealed that there are no effects of gene mutations on translation of the product protein.
