الساركوفين هو مركب طبيعي مستخلص من الشعاب المرجانية المتواجدة في البحر الأحمر يعرف باسم الساركوفيتون.
و لمركب الساركوفين العديد من الأهميات البيولوجية و منها أنه مثبط لكثير من الخلايا السرطانية.
و على الرغم من ذلك فإن تفاعل الساركوفين مع الحامض النووي لم يدرس بعد، لذلك فقد كان هدف هذا البحث دراسة إمكانية ارتباط الساركوفين مع الحامض النووي الديوكسي ريبوزي.
و من خلال هذا البحث تم دراسة تأثير الساركوفين على الحامض النووي الديوكسي ريبوزي معمليا خارج جسم الكائن الحي باستخدام تركيزات مختلفة من الساركوفين تم إضافتها إلى الحامض النووي, أتبعت بقياسات آليات الفصل الكهربائي الأفقي و قياسات الأشعة تحت الحمراء.
و قد أظهرت نتائج هذه الدراسة أن مركب الساركوفين ذو السمية الطبيعية له قابلية كبيرة للارتباط بالحامض النووي الديوكسي ريبوزي.
و هذه النتائج سوف تساعد الباحثين على فهم ميكانيكية عمل الساركوفين كمثبط للخلايا السرطانية.
Abstract EN
Sarcophine is a natural product isolated from the Red Sea soft coral Sarcophyton glaucum and other sarcophyton species.
It has a cembranolide diterpene structure.
Sarcophine was shown to possess numerous biological activities including a potent antitumorigenicity.
The interaction between sarcophine and nucleic acids was not studied yet.
This warranted a study of its potential chemical affinity to DNA.
In the current investigation, the in-vitro effects of sarcophine on DNA were studied.
Different concentrations of sarcophine were added to DNA followed by the electrophoretic mobility shift assay and the UV-spectrophotometric measurements.
The results of this study showed that the natural toxin sarcophine has a high binding affinity towards double stranded DNA.
These findings may help in understanding the mechanism of the anti-tumorigenic activity of sarcophine.
