إن الاهتمام بتأثير المليتين في الآونة الأخيرة كأحد المكونات المنشطة للمناعة في سم النحل نتج عنه دراسات للبحث عن العلاقة بينه و بين الأمراض المستعصية و الأورام.
و قد هدفت الدراسة الحالية إلى محاولة معرفة التأثير المنشط للمناعة للمعالجة بالمليتين على نمو الورم المستحدث في الفئران باستخدام خلايا إيرلش الورمية و ذلك من خلال دراسات خارج الجسم الحي أو داخله, هذا بالإضافة إلى محاولة التعرف على ميكانيكية العمل المحتملة لهذا التأثير.
و قد أظهرت النتائج أن حقن المليتين تحت الجلد في الفئران بجرعات (0.5, 5 أو 50 ميكروجم / 100 جم من وزن الجسم) يوما بعد يوم لمدة 4 أسابيع و ذلك قبل حقن خلايا إيرلش الورمية (1×610 خلية) في التجويف البريتوني للجسم قد تسبب في زيادة ملحوظة في أعداد الخلايا الليمفاوية في الغدة التيموسية و نخاع العظم و كذلك خلايا الرشيح البريتوني, كما أدى إلى تنشيط الوظيفة البلعمية و وظيفة الخلايا التائية و البائية و كذلك وظيفة كل من الكبد و الكلية.
أما عند قياس نشاط الورم, فإن المعالجة بالمليتين قد قللت من تكاثر خلايا إيرلش الورمية داخل الجسم, كما قللت من حيوية هذه الخلايا خارج الجسم.
بالإضافة إلى ذلك, فقد تسببت المعالجة بالمليتين في نقص حجم الورم الصلب نقصا معنويا و ذلك عند قياسه مورفولوجيا و فحصه نسيجيا.
و تخلص هذه الدراسة إلى أن قدرة المليتين المثبطة لنمو الورم ترجع لآلية غير مباشرة تعتمد على تنشيط الاستجابة المناعية الخلوية و السائلة.
Abstract EN
Recent interest in the action of melittin, as an immunostimulator, should result in research of its intimate relationship with a wide range of incurable degenerative and malignant diseases.
The objective of the present study was to investigate the immunostimulatory effect of melittin pre-treatment against Ehrlich tumor growth in mice both in vivo and in vitro and the possible mechanism(s) of antitumor activity.
Subcutaneous injection of melittin with doses (0.5, 5 and 50 ?g/100 g b.wt, every other day for consecutive 4 weeks) before inoculation of Ehrlich ascite tumor (EAT, 1x106 cells) to mice elicited a significant increase in the cellularity of both thymus and bone marrow lymphocytes as well as peritoneal macrophages.
Melittin pre-treatment activated the phagocytic function of macrophages, as well as T- and B-cell functions, and recovered both liver and kidney functions.
Melittin pre-treatment also decreased the proliferation of tumor cells incubated in the peritoneal cavity of mice, and decreased their viability percentage in vitro.
When measured morphometrically and examined histologically, the solid Ehrlich tumor was significantly reduced in size.
The present study proposed that melittin has indirect mechanism of tumor growth inhibition that is based on activation of both cell-mediated and humoral immune responses.
