The present study characterized the Plectropomus maculates fish sample obtained from Yanbu coast on the Red Sea in Saudi Arabia by morphological parameters and by a fragment of 200 bp represented the 16S rRNA gene for use in differentiating Saudi domestic species.
Blast alignment of 16S rRNA gene with NCBI GenBank revealed three P.
maculates accessions of related samples Alignment of sequence from a given species revealed similarity ranged from 97 to 98%.
The alignment allowed the development and analysis of consensus sequences for Plectropomus maculates sample and comparison with limited sequences in public database GenBank.
The results also showed similarity with the other sample species in the GenBank with percentages ranged from 84 to 95%.
P.
maculates sample revealed 11 positional differences with base-pair substitutions.
They revealed four transversions interchanges of purinepyrimidine nucleotide, one adenine was also changed to thymaine and other three transitions were existed in the sample under study.
The phylogenetic dendrogram divided all named GenBank P.
maculates species into two main discrete clusters, whereas the fish sample under study formed a phylogenetically distinct cluster and was separated from the three P.
areolatus and the other three P.
maculatus existed in GenBank.
The obtained result evidently indicated diversity with unique characteristics of the P.
maculates fish sample collected from Yanbu coast on the Red Sea in Saudi Arabia over all the other Plectropomus samples established in NCBI GenBank.
The obtained results confirmed that the geographical attitude of the environment such as pH and temperature may decrease the similarity from 99 or 100% to less percentage with other samples in the GenBank.
However, P.
maculates of Yanbu coast showed convergence between P.
maculate and P.
maculates samples in the NCBI GenBank.
