English Abstract
Bacterium Acinitobacter calcoaceticus was isolated from agriculture soil of Al-mafraq governate - Jordan.
It was identified by using (16S ribosomal RNA).
This study was conducted to evaluate the ability of Acinitobacter calcoaceticus to degrade chlorobenzoic acid compounds especially 3-CBA and to evaluate the optimum condition for degradation.
Acinitobacter calcoaceticus was found to use the chlorobenzoate compounds as alternative carbon and energy source.
The percentages of degradation of the chrobenzoates 2-CBA, 3-CBA, 4-CBA and 3,4-dCBA at the concentration 2-mM were found after 80hr of incubation was 55%, 73%, 60.1% a and 54.9% respectively.
The order of initial rates were 30.3μM/h for (2-CBA), 20.6μM/h for (4-CBA), 10μM/h for (3,4-dCBA) and 55μM/h for (3-CBA).
The best percentage of degradation of 3-CBA compound were obtained with 3mM and 4mM substrate concentration and it was 73%.
Adaptation of the cells on 3-CBA for 48 hours and cells starvation for 24 hours and 48 hours increased significantly the initial degradation rate.
The carbon sources increased the cell biomass.
However, all carbon sources used reduced the biodegradation.
Nitrogen source reduced the degradation activity of the 3-CBA as well as the chlorine release from it.
Best inoculum size was 1.5xml.
Best agitation rate was 150RPM.
The experiments were included temperature and ph.
The preferential temperature and pH for the strain were 37ºC and ph.
