English Abstract

The study is aimed to measure the activity of arginase enzyme and purifying it from sera of females with uterine fibroid.

In this study, specimens were collected during the period from 1st Dec.

2013 until the end of May 2014 at Obstetrics and Gynecology department of two hospitals in Baghdad city (Baghdad teaching hospital, and Al-Yarmuk teaching hospital) consisted of (50) females with uterine fibroid and (30) healthy females being classed as a comparison group and their ages ranged between (30-55) years for two groups.

The results showed that there was a highly significant increase (P≤ 0.001) in the arginase activity in sera of females with uterine fibroid (7.99 ± 0.23) I.U/L, while the mean of the activity was (0.52 ± 0.02) I.U/L in the healthy females.

Moreover, the study included purification by chromatography of gel filtration using Sephadex G-50 .The arginase isoenzymes were isolation by chromatography of ion exchange by using DEAE Cellulose A50, and four isoenzymes were obtained.

Also the study included measurement of some biochemical parameters in the sera of females with uterine fibroid which included estimation the level of (adiponcetin, progesterone, estradiol, prolactin) and some anti-oxidants (malondialdehyde, peroxynitrite) and compared with the healthy females.

The patients showed a highly significant increase (p≤0.001) in the level of (progesterone, estradiol, prolactin, malondialdehyde, peroxynitrite), while they showed a highly significant decrease (p≤0.001) in the level of adiponectin compared with healthy females. The correlation coefficient(r) between the enzyme level and other parameters calculated with regression plot showed a positive correlation between arginase with (progesterone, prolactin, malondialdehyde, peroxynitrite) in the uterine fibroid females, While it was noted that there was a negative correlation between arginase (adiponcetin, estradiol) in the uterine fibroid females.

On other hand, it was notested in this study that there was a positive relation between adiponectin with (estradiol, peroxynitrite) and a negative relation between (progesterone, prolactin, malondialdehyde) in the uterine fibroid females
