Objective: the Purpose of this study was to evaluate in vitro the apical microleakage of four different root canal sealers (ZOE , Realseal , EndoREZ , Apexit plus ) using different obturation methods (single cone , lateral condensation) .

Materials and Methods: The study was carried out using 80 newly extracted single – canal permanent anterior teeth distributed into 8 groups (each 10 teeth ).

All teeth were cleaned and shaped using a set of ProTaper rotary instruments.

Groups 1 and 2 were filled with Realseal and Resilon , Groups 3 and 4 filled by EndoREZ and GP, Groups 5 and 6 filled by Apexit plus and GP , Groups 7and 8were filled with Zinc Oxide and Eugenol (ZOE) and GP and considered as control Groups ,.

Groups 1,3,5 and 7 were filled using single cone technique while Groups 2,4,6 and 8 were filled using lateral condensation technique .

After the specimens were stored in 100% humidity at 37 degrees C for 1 wk, the roots were covered with two layers of nail polish and The apical leakage was measured using the dye penetration technique using 2% methylene blue as a tracer for 24 h, followed by longitudinal sectioning of the specimens .

Results: Data were subjected to analysis.

P<0.05 was considered as significant.

When lateral condensation technique was used Kruskal-Wallis test showed no significant differences between ZOE, Realseal , EndoREZ, Apexit plus root canal sealers ( p>0.05).

Kruskal-Wallis test showed significant differences between ZOE , Realseal , EndoREZ, Apexit plus root canal sealers when using single cone technique.

However, Resilon and Realseal sealer showed the least apical leakage (p<0.05).

Kruskal-Wallis showed no significant differences were found between lateral condensation technique and single cone technique when using Realseal , EndoREZ, Apexit plus root canal sealers (p >0.05).

Conclusion: There was no significant difference in apical leakage between single cone technique and lateral condensation technique when Realseal, EndoREZ , Apexit plus root canal sealers were used .

When lateral condensation technique was used: there was no significant difference in apical leakage between All root canal sealers used
