Abstract EN

The beetles Tribolium confusum is the most important economical pests in the world that have problem in taxonmy because it resembles Tribolium castaneum and it is difficult to distinguish between these two species in the immature stages and adult stage.

Therefore, the research is to study their morphological and molecular characteristics.

In this research, PCR (Polymerase chain Reaction) and PCR-RFLP was used for corroboration of morphological classification of T.

confusum at the molecular level.

The adult insects have been classified depending on the taxonomy, the DNA was extracted from the insects, and a technique PCR was performed with the 28S rDNA using specific primers (D2-3665F/D5-4749R) to T.

confusum.

After the 28S rDNA gene was amplified, the electrophoretic PCR products shown that length of fragment 28S rDNA gene was 1072 bp to all members of T.

confusum.

(larval stages and adult stage).

Corroboration of the Morphological classification of T.

confusum, PCR-RFLP was used, and the electrophoretic PCR products showed 3 bands with length ٥٠٠-٤٥٠-٢٧٥ base pair.

Thus the corroboration of morphological classification of T.

confusum at molecular level was achieved.
