Polymorphism in promoters of TLR-2 gene in Iraqi patients with tuberculosis

Despite the cxistcncc of Tubcrculosis control programs globally, TB romains onc of the major public hcalth problems worldwide.

It is onc of the grcatest challenges, which face hcalth systems in the 21“ ccntury.

A dcfcct in the genes of the immunc response is the most plausible explanation for susccptibility of some individuals and résistance of others to TB.

TLR2(CD282) is onc of the toll-like reccptors and plays a rôle in the immunc system.

TLR2 is présent on the surface of certain cells and it distinguishes forcign bodics, also it have rôle in passing of adéquate signais to the cells of the immunc system.

The main goal of this study is to produce molecular analysis for promoter of TLR-2 genes in Iraqi patients writh TB.

Sevcnty-four blood spccimcns were collcctcd from 74 patients in the Institutc of the Tubcrculosis and Ch est Disease-Basra city's, blood spccimcns also collcctcd from 74 hcalthy individuals as control.

Extracted DNA was amplified using set of spécifié primers for promoter régions TLR-2 genc; amplified amplicons were sequeneed and analyzcd.

Gcnotyping of TLR-2 gcnc promoter rcvcalcd that out of 74 found with TB, 38 (51.3%) patients found to possess C allele (T—»C), while 36 (48.7%) possessed T allele.

C allele présents only in heterozygous statc in Iraqi society, and closcly corrclatcd with increasing susccptibility of some individuals to TB.
