Detection one of DNA repair gene for lung cancer in sample of Iraqi patients

The DNA scquencc can bc changcd and damage by cnvironmcntal agents such as mutagcnic chcmicals and certain types of radiation.

The correction of DNA scquencc errors in ail types of cells is important for survival.

DNA gcnc repair is an important way to repair this damage.

Onc of DNA repair genes is the Ref-1 gcnc, Howevcr Ref-1 gcnc like other genes contains mutations which are found in différent sites.

In présent study, whole blood was collcctcd from 20 individuals distributcd to two groups:- Tcn lung cancer patients from various provinces and both gender and différent age groups and Ten volunteers from hcalthy pcoplc as control group.

DNA was extracted with RcliaPrep Blood gDNA Miniprcp System and the results of Ref-1 by PCR gave positive for ail samplcs.

then analysis PCR product of 5 samplcs (patients and normal ), results compared with scqucncing of original Rcf-lgene in NCBI showed similarity between scqucnccs of normal samplcs, while there wrerc différences in the scqucnccs of samplcs lung cancer patients.

Results of scqucncc analysis Ref-1 showred that were 11 mutation in 5 patients samplcs writh gcnc Ref-1 and showr single mutation in some samplcs .Mutations Icad to changes in the gcnctic code and in amino acids that is important reasons of an incrcasc in the gcnc expression changes in Ref-1 gcnc.
