β-galactosidase enzyme was extracted from the udder of cows by using six methods, and purified using four different methods.

It was found that the method of 0.2 mol/L of acetic acid+ 0.2 mol/L ascorbic acid pH 5 was the best and gave the highest specific activity of crude enzyme in comparison with other used methods.

The use of ethanol50%was found the best way of purification.

Using gel filtration Sephacryl S-200 (second step), allowed to have purificationfactor6.53 times and the yields was 89.36%.
