الهدف من هذه الدراسة هو محاولة إثبات صحة مجموعة (Kit) تجارية لقياس هرمون الأديبونكتين في الفئران لاستخدامها في القطط.
الأديبونكتين هرمون يفرز من الخلايا الدهنية و له علاقة مباشرة مع البدانة و مقاومة الأنسولين في الإنسان و الفئران.
دراسة هذا الهرمون في القطط سوف تؤدي إلى تحسين طرق فحص و علاج السكر في القطط.
تم قياس الاختلافات داخل الاختبار الواحد و بين الاختبارات و بعضها و كذلك تنوع و خطية و موازاة قدرة اختبار ELISA على استخدامه في أمصال القطط.
بروتين الأديبونكتين القطط تم فصله عن البروتينات الأخرى عن طريق الاستبعاد الحجمي بواسطة الفصل اللو في chromatorephy ثم تبعه بطريقة SDS–PAGE المختزل.
قياس معامل الاختلاف داخل الاختبار الواحد كانت 3.7 % أما بين الاختبارات و بعضها فكان معامل الاختلاف 6.5 %.
و قد أوضحت النتائج خطية مقبولة مع استخدام المنحنى المعياري (± 0.02) R2 of 0.97.
كان متوسط الاستعادة لهرمون الأديبونكتين المصافي العينات 84 % (+15).
المعاولات المستخدمة لإثبات صحة اختبار ELISA كانت مقبولة لاستخدام هذه المجموعة التجارية لاستخدامها في القطط.
Abstract EN
The goal of this study was to validate a commercial mouse / rat adiponectin ELISA kit for use in cats.
Adiponectin is a hormone secreted from adipocytes closely associated with obesity and insulin resistance in humans and mice.
Studying of this hormone in cats may improve methods for screening and treating feline diabetes.
Intraassay and inter-assay variation, linearity, parallelism, and recovery of the ELISA were measured using feline serum.
Feline adiponectin was separated from other serum proteins using size-exclusion gel chromatography, followed by reducing SDS gel PAGE electrophoresis and used to confirm binding with the ELISA.
The Intra-assay and interassay coefficients of variation were 3.7 % and 6.5 %, respectively.
Samples showed acceptable linearity with an average R2 of 0.97 ( ± 0.02) and were parallel to the standard curve.
The average recovery of mouse adiponectin spiked into samples was 84 % ( ± 15).
The parameters for validation of the ELISA assay are acceptable and support the use of this assay for detection of feline adiponectin in serum.
