خلفية البحث : سرطان عنق الرحم مرض يهدد حياة الكثير من النساء حول العالم.
و بالرغم من تعدد علاجات سرطان عنق الرحم المستخدمة حاليا إلا أنها تستحدث العديد من الآثار الجانبية.
لذلك تبذل الجهود الحثيثة لاكتشاف علاجات من مصادر طبيعية آمنة كالنباتات مثلا نبات القريص ( Urtica pilulifera ) كأحد هذه النباتات تم تسجيل فاعليته ضد عدة أنواع من السرطان لكن آلية عمله لم يتم التحقق منها بشكل تام بعد.
أهداف البحث : تقييم النشاط المضاد للسرطان الذي تمارسه مركبات كيميائية نباتية مستخلصة من أوراق نبات القريص ضد خلايا سرطان عنق الرحم ( Hela ).
منهجية البحث : تم تقييم النشاط المضاد للسرطان لمستخلصات خام من أوراق نبات القريص ( الهكسان - الميثانول - الماء ) من خلال تحديد التأثير السمي على خلايا Hela بواسطة اختبار MTT تلى ذلك عملية تجزئة المستخلص الخام الأقوى و تقييم نشاط الأجزاء أيضا بواسطة اختبار MTT بالإضافة إلى إجراء تحليل كروماتوغرافيا الغاز- مطياف الكتلة للمستخلص الخام و كل الأجزاء الجزء الأكثر فعالية تم اختباره أكثر من خلال إجراء اختبار trypan blue لتقييم قدرته على قتل الخلايا.
أُجري تحليل ( Western blot ) للكشف عن آلية العمل التي اتبعها الجزء عن طريق فحص أي تغيرات في مستويات البروتينات1-P21، P53، PARP.Caspase-9، BCL-2.
نتائج البحث : أظهرت نتائجنا أن ug/ml45 ) من المستخلص الهكساني للقريص أظهر سنية قوية ضد خلايا Hela الجزء الرابع من هذا المستخلص أظهر سمية أقوى ضد نفس الخلايا بقيمة ( IC50 ) تساوي ( 40ug/ml ).
الجزء الرابع كان يضم عدد من المركبات والتي أهمها الفيتول الذي وُجد في الأجزاء الفعالة فقط.
بالإضافة إلى ذلك، الجزء الرابع استحث نشاطه المضاد للسرطان من خلال آليتين وقف دورة الخلية و الموت الخلوي المبرمج.
الخلاصة و التوصيات : المركبات الكيميائية النباتية المستخلصة من ورق نبات القريص ذات نشاط قوي ضد خلايا Hela.
English Abstract
Background: Cervical cancer is a life-threatening disease for many women around the world.
Although many treatments are currently used to treat cervical cancer, they contribute to induce many side effects.
Therefore, efforts are being made to discover treatments from safe natural sources such as plants.
Urtica pilulifera as one of these plants has reported to be effective against several types of cancer but whose mechanism of action has not been fully verified yet.
Objective: To evaluate the anticancer activity exerted by phytochemicals extracted from U.
pilulifera leaves against cervical cancer cell line (Hela).
Methodology: The anticancer effect of hexane, methanol and water crude extracts of U.
pilulifera leaves was assessed through determining cytotoxic effect on Hela cells by MTT assay.
This was followed by fractionating the potent extract and assessing fractions activities by MTT assay too, in addition to performing GC-MS analysis for crude extract and all fractions.
The most effective fraction was further tested by trypan blue assay to evaluate its ability to induce cell death.
Western blot analysis was carried out to detect the mechanism of action by which this fraction followed by cheking any alterations in P21, P53, PARP-1, Caspase-9 and BCL-2 proteins levels.
Results: Our results showed that 45 µg/ml of U.
pilulifera leaves hexanic extract exhibited a potent cytotoxicity against Hela cells.
Of this extract, fraction 4 showed most potent cytotxicty againist the same cell line with 40 µg/ml IC50.
Fraction 4 contained several compounds, the most important of which is phytol, which was found in active fractions only.
Furthermore, fraction 4 induced its anticancer activity through two mechanisms of action, cell cycle arrest and apoptosis.
Conclusion: Phytochemicals extracted from U.
pilulifera leaves have a potent anticancer activity againist Hela cell line.
