English Abstract

This study was conducted on eleven cultivars of date palm varieties to analyze the chloroplast DNA (cpDNA) genetic variation.

cpDNA was extracted with sufficient quantities and high purity ranged between 1.7 and 1.9 , and specific primers were selected to amplified three regions from cpDNA.

One band was result from all varieties with molecular weight 400bp after performing the PCR reaction using trnL-trnF region primer and 800bp and 512bp with matK and rpoB genes, respectively.

RFLP-PCR technique was used to reach the goal, two restriction enzymes were selected in this technique.

Three bands were result from all varieties with molecular weight 390 bp, 269 bp , and 141 bp after treatment pcr products of matK gene primer with Taq1 restriction enzyme.As for trnLtrnF region and rpoB gene primers did not appear any result with this enzyme.

With EcoRV restriction enzyme two bands were result from all varieties with molecular weight 423 bp and 377 bp except Sugary and Maktoum, thus, this enzyme was able to distinguish these two varieties from the other varieties, but it was not able to distinguish between them.

Likewise,as for trnL-trnF region and rpoB gene primers did not appear any result with this enzyme.
