The objectives of this study were to isolate, molecular characterize and screen of new local isolates with the potentially product agents.

Screening revealed that 35 out of 103 sample of crude cow milk (34%) were Lactococcus lactis subsp.

lactis.

Characterization of these selected isolates using conventional method, API technique and further confirmation was achieved by PCR amplifying.

All isolated exhibited marked activity against five pathogenic bacterial strains (Bacillus subtilis (ATCC), Staphylococcus aureus (ATCC 43306), Escherichia coli (ATCC 25922), Pseudomonas aeruginosa (ATCC 27853) and (Clostridium perfringens), using agar well diffusion method.

Isolate 58 and 63 showed the highest activity (20mm) and (15 mm) respectively, against Gram positive bacteria B.

subtilis and S.

aureus.

Where as isolate 70 showed the highest activity (13 mm) against gram negative bacteria P.

aeruginosa, generally all aforementioned isolates were active against Gram positive and Gram negative bacteria.

Isolate 46 exhibited its maximum activity (30mm) against Clostridium perfringens at pH 2.5, isolate 42 had its highest activity (20mm) against Clostridium perfringens at temperature -4°C.

Under different UV light wave strength all isolate showed low or no activity against the different pathogenic bacteria induced in this study.

The antibacterial activity of the isolate 58 was compared with four standard commercial antibiotics ampicilin (AS), cephalexin (PR), cefotaxine (CF), cloxacillin (CX).

Showed higher activities against B.

subtilis (20mm) and (9mm) against S.

aureus more than commercial antibiotic.

In conclusion these results advocate that isolate 58 could be as a promising isolate against the Gram positive and Gram negative pathogenic bacteria with higher antibacterial activity.
