Radioimmuno assay was used to measure the levels of Beta-2-microglobulin (b2M) in the sera of three women groups, group I consisted of (28) post-menopausal women with malignant breast tumor, group II consisted of (22) pre-menopausal women with malignant breast tumor and group III consisted of (25) pre-menopausal benign breast tumors.
A fourth group of (20) healthy women was also included as control.
The results showed significant differences between each group of cancer patients and the control group (P < 0.001).
The binding of 125I-b2M to its receptors in the cytosolic fraction of breast tumor homogenates was studied by using the radioimmuno assay technique with our modification for tissue ( radioreceptor assay).
The receptors for b2M were isolated from malignant post-menopausal homogenate using gel filtration chromatography technique.
Two forms of b2M receptors were found (protein I and protein II), which have a molecular weight 52.8 and 29 KD respectively.
The isolation process caused an increase in the binding affinity whereas the purification fold was 6 for protein I and 4.5 for protein II.
Time course and kinetic studies on the binding of 125I-b2M to its crude and isolated receptors were carried out at different temperatures.
The binding reaction is a time and temperature dependent process and follows the second order rate law in all studied samples.
The thermodynamic parameters of the standard state (DHo, DSo and DGo) and the transition state (DH*, DS* and DG*) and activation energy (Ea) were determined for 125I-b2M binding to its receptors at different temperatures.
These parameters were used to characterize the binding reaction that was found to be endothermic and spontaneous in all studied groups.
The spectroscopic characterization of b2M and (125I-b2M/receptor) complex was studied using ultraviolet spectroscopy.
Factors affecting the absorption spectra of the two species such as medium pH and solvent polarity were investigated.
The pKa values of histidine and tyrosine residues and the percentages for the presence of these residues on the surface of the two species (b2M and 125I-b2M / receptor complex) were studied using spectroscopic pH titration.
