Potential production/ extraction of bioactive molecules was investigated in Taxus baccata Var.

Dovastoniana and from microorganisms isolated from associated biotopes.

Taxus baccata needles were grounded and extracted using acidic water.

Two procedures were used in order to extract taxoids.

The different extracts were tested for their polyphenols content and their antioxidant capacity.

The first procedure showed that the ethyl acetate extract exhibits the highest amount of polyphenols and the lowest antioxidant activity.

The extracts obtained from the second procedure were analyzed for their polyphenols content, their antioxidant power and the DPPH-scavenging activity.

The ethyl acetate plant extracts showed high polyphenols content, ranged between 20 to 30 mg/100 mg of extract and they exhibited DPPH scavenging activity.

The extracts were analyzed with HPLC/ESI-MS in order to screen the presence of taxanes.

The chloroform extract proved the presence of several taxane-like molecules, mainly paclitaxel and 10- deacetylbaccatin III.

In parallel, bacteria from three Taxus baccata related biotopes (soil, needles and bark) were isolated and screened for their ability to produce taxanes-like molecules based on the analysis of the bacterial extract on TLC.

Out of 166 isolates, nineteen were selected for further analysis represented by testing their antioxidant capacity and their polyphenols content.

The isolates were subjected to RFLP, protein profile and carbohydrate test for taxonomic characterization.

In addition, these bacteria were tested for their biochemical properties such as catalse activity, oxidase activity and Gram staining.

Phylogenetic analysis was done by sequencing a fragment of 757 bp using the universal primers fd1 and RS 16.

The obtained results showed that L4 bacterium belongs to the genus Pantoea.

The production of taxane-like molecules was assessed by HPLC/MS by analyzing the extract from the free cell culture of two isolates: L2B and L4.

The results showed the ability of these isolates to produce taxane-like molecules using tyrosine as substrate.

The HPLC/MS detected the production of pacliaxel and 10 deacylbaccatin III.

These results suggest an economically affordable and efficient process for the taxane-like molecules production.
