Background : There is a two way relationship between diabetes and periodontitis.
AGE accumulates in the tissues by aging or due to hyperglycemic state.
AGE affect the periodontal tissues through binding with RAGE specific receptor and stimulate a cascade of inflammatory and immune reactions leading to tissue destruction.
There is a soluble form of RAGE called sRAGE present normally in the body and acts as a competitive inhibitor to RAGE by binding to its ligands and prevent tissue destruction.
Aim : Was to assess the frequency of Expression of mRNA of the soluble Receptor of Advanced Glycosylated End products (sRAGE) in gingival tissues of type 2 diabetic patients affected by chronic periodontitis by the use of PCR.
Method : -The subjects of the study were 36 patients grouped into three groups G I systemically free affected by chronic periodontitis as a control group (n = 12), G II well controlled diabetic subjects affected by type 2 diabetes and chronic periodontitis (n = 12) and G III poorly controlled diabetic patients affected by type 2 diabetes and chronic periodontitis (n = 12).
Gingival biopsies were collected during surgical periodontal therapy of the subjects and stored at (- 70) to be used for mRNA extraction and subsequent RT–PCR detection of the sRAGE.
Results : The present study showed that the expression of sRAGE was increased in well controlled diabetics and in non diabetics with less scores and measurements of clinical parameters.
sRAGE mRNA in the present study was negatively expressed in 83.33 % of the poorly controlled diabetics and in patients presented with more periodontal tissue destruction.
Conclusion : sRAGE was reported to be associated with non diabetic states and with glycemic control of type 2 diabetes and inversely associated with components of the metabolic syndrome.
