تهدف الدراسة إلى كبح الأنزيم β–glucosidase الذي يثور شلبا على آلية عمل الأنزيم Exo-β–glucosidase و لهذا الغرض تم تنمية الفطر Trichoderme viride على المخلفات الورقية و الذي يمتلك القدره الفائقه على إنتاج المعقد الأنزيمي المتعدد Cellulase ذو القابلية العالية على تهشيم السلاسل السيليلوزية.
تم تثبيط فعالية الأنزيم β-glucosidase باستعمال الماده المثبطه D –glucano- б–lactonالتي نجحت في كبح فعالية الأنزيم β-glucosidase خلال دقيقه واحده حوالي ٩٠ % و عند تركيز 0.5 ملغم / مل، لوحظ ارتفاع فعالية الانزيم Exo–1, 4-β–glucosidase حيث بلغت 12 وحده / مل.
تم تحديد الظروف المثلى من درجة حراره و الأس الهيدروجيني و مدة عمليات التخمير للانزيم Exo-1, 4-β – glucosidase ، و كانت (4 م، 4.8، 8 يوم) على التوالي والتي اسهمت في مضاعفة فعالية الأنزيم بمقدار ثلاثة مرات و اختزال مدة عمليات التخمير 48 ساعة.
Abstract EN
The aim of this work was to inhibition of the enzyme β–glucosidase for the production of the enzyme Exo-1, 4-β-glucosidase that have the power for analysis of cellulose to glucose.
The using of the inhibition substance D- glucano - δ - Lacton success to inhibition of β–glucosidase at the concentration 0.45 mg/mL.
at 1 minute. The optimum circumstances temperature, pH, and time of fermentation were (45 min., 4.8, 8 day).
