من خلال الدراسة الحالية تم عزل الكانديدا البكانس من التجويف الفمي للإنسان و تم تعريفها بالطرق التقليدية ثم تم تأكيد التعريف بواسطة PCR و بعد ذلك تم تعريض الكانديدا للأشعة الفوق بنفسجية لمدة 30 دقيقة على مسافة 15 سم ثم تم عزل المادة النووية و إجراء PCR ثم التتابع الجيني للجين ساب 2 باستخدام نوعين من البرامير الخاص بهذا الجين لتأكيد تأثير الأشعة الفوق بنفسجية على جين ساب 2 التي أثبتت الدراسة حدوث تغير في التتابع الجيني للكانديدا البكانس المعرضة بنسبة 5% عن العينة التي لم يتم تعريضها.

Abstract EN

Extracellular proteolytic activity play a great role in Candida albicans pathogenicity and is produced by a family of 10 secreted aspartyl proteinase (sap) genes.

Sap2 is the most important member of them.

The present work was done to study the influence of ultraviolet on sap2 gene.

Isolation of genomic DNA for PCR analysis and sequencing to detect sap2 gene in radiated and non radiated Candida albicans were done.

The results reveal that, sap2 gene sequencing was changed after UV.

The change in some location of the gene lead to decreasing its ability to secrete Aspartyl proteinase.
