Arabic Abstract
تشير نتائج الدراسة أن مستخلص الإنزيم من نباتات النعنع و الحصلبان و الزعتر و ورق الزيتون لها نشاطية لتحطيم البروتين عند درجة حموضة (3 إلى 7) و درجة حرارة مثلى عند (35 إلى 45)، و عند دراسة المجموعات المتأينة حول الموقع النشط للأنزيم من خلال Dixon-Webb’s plot وجد أن aspartic acid glutamic acid and cysteine موجودة في أو حول الموقع النشط للأنزيم و هذه الأنزيمات تنتمي للإنزيمات الحمضية و القاعدية، الدراسات الحركية للأنزيمات (km, vmax, vmax / km) وجد أن الكازين أفضل مادة متفاعلة مع الإنزيم في نبات الحصلبان ثم يليها نبات النعنع ثم الزعتر و أخيرا ورق الزيتون و وجد أن بياض البيض أفضل مادة متفاعلة مع الإنزيم في نبات النعنع ثم نبات الزعتر ثم ورق الزيتون و أثبتت دراسة km ان الكازين له فعالية كبيرة في نبات الحصلبان ثم الزعتر ثم النعنع و أخيرا ورق الزيتون.
English Abstract
Our result illustrate that the crude enzyme extracts of Mentha piperita, Rosmarinus officinalis, Thymus capitatus and Olea europaea have a proteolytic activity within optimum pH (3-7) and optimum temperature (35-45 oC) .
Study of the ionizable groups in or around active site of these proteases by Dixon-Webb’s plot reveals the occurrences or the presence of aspartic acid, glutamic acid and cysteine amino acids in or around the active site , interestingly , these proteases may be belonged to the acid proteases and neutral proteases.
Enzymatic kinetics studies (km, vmax, vmax / km) indicated that casein is the most specific substrate for proteases of R.
officinalis followed by M.
piperita, T.
capitatus and O.
europaea, while egg albumin is the best substrate of M.
piperita followed by T.
capitatus, O.
europaea.
In contrast, results of km values indicated that casein has high affinity for proteases of R.
officinalis, T.
capitatus, M.
piperita and O.
europaea
