English Abstract
The proteolytic activity in crude extracts of pumpkin and pomegranate fruits was measured with albumin, egg albumin and casein as substrates.
The kinetic parameters (Vmax) and (Km) were investigated using the different substrates and revealed a broad specificity from both samples.
Both crude extracts appeared to be active over a wide pH range with optimum activity at pH 7 and at a temperature of 40 ˚C.
The enzymes from pumpkin were purified 6.9 fold and from pomegranate 36.5 fold.
The enzyme from both crude extracts of pumpkin and pomegranate may be a cysteine proteases as indicated by Dixon and Webb’s plot of Log10(Vmax / Km) against pH, which revealed an apparent pKa1 and pKa2 of 6.3 and 7.6, and 5.6 and 6.8, respectively, indicating that histidine and cysteine amino acid residues may be essential for enzyme activity.
