تعتبر الفطريات الجلدية من الفطريات الخيطية و التي تنتمي لجنس و عائله التريكوفيتون، الإبيدير مفيتون و الميكروسبوروم.
إن الإصابة بالفطريات الجلدية لا تقتصر على الحيوانات فقط و لكنها تنتقل بسهولة للإنسان.
أثناء فترة العدوى الفطرية تكمن خطورتها في إفراز الفطر لمجموعة من الإنزيمات التي تساعد الفطر على اختراق نسيج العائل المصاب بالفطر من أهم هذه الإنزيمات تلك المحللة للبروتين والتي تحتوي على الإنزيم الفطري المحلل من الأنواع (MEP1-5) و التي تعتبر من أهم عوامل الضرورة التي يمتلكها الفطر.
في هذه الدراسة تم عزل و تصنيف الفطريات الجلدية من عينات من حالات مصابة و كانت من نوع التريكوفيتون فيركوزوم و تم فحص المعزولات لوجود الجين المسئول عن الإنزيم المحلل الفطري باستخدام تفاعل البلمرة المتسلسل و قد أظهرت النتائج إن معدل تواجد الجينات المسئولة عن الإنزيم المحلل الفطري (MEP1-4) قد ظهرت في 10 % من العينات المعزولة و المصنفة، بينما في حالة (MEP5) قد ظهرت في 20 % من العينات المعزولة و المصنفة.
تعتبر هذه النتائج هي البيانات الأولى التي أجريت لفحص مدى انتشار هذه الجينات في المعزولات المحلية للنوع تريكوفيتون فيريكوزوم و على الرغم من ذلك لا نزال نحتاج إلى مزيد من الدراسات و البحث للتحقق من الدور الدقيق التي تلعبه هذه الإنزيمات في مساعدة العدوى بالفطريات الجلدية في مصر.
و علاوة على ذلك، لا بد من تحليل تسلسل الحمض النووي للتحقق من الاختلاف الوراثي بين السلالات المعزولة بشكل جماعي، هذه الدراسة سوف تكون هذه الدراسة حجر الأساس من أجل فهم العدوى بالفطريات الجلدية و عوامل ضرورتها مما يساعد في تطوير طرق التدخل العلاجي المناسبة.
Abstract EN
Dermatophytes are filamentous fungi that belong to genera Trichophyton, Epidermophyton and Microsporium.
Dermatophytes not only can cause a variety of dermatophytosis in animals but also can transmit to humans.
During the course of infection, the severity of infection requires the release of specific enzymes that facilitate the penetration into the host tissue.
Several proteases including fungalysins (MEP1-5) have been considered as a potential virulence factors.
In the present study dermatophyte was isolated and identified from infected samples and then samples were examined for the presence of these genes (fungalysins) by Polymerase Chain Reaction (PCR).
The data collected from this study showed that, the incidence of MEP1-4 was only 10 % of the screened samples, while it was 20 % for MEP5.
Although, to our knowledge these findings considered as the first data carried out to screen the incidence of these genes in dermatophytes (Trichophyton verrucosum) isolated from local strains, however, further investigations are still required to verify their precise role in dermatophytes infection in Egypt.
Moreover, sequence analysis is required as well to verify the genetic difference between the isolated strains.
Collectively, this will be the basal block in order to understand the dermatophytes infection virulence attribute and helps in developing of the appropriate therapeutic intervention.
