English Abstract
Phenoloxidase (PO) activity was studied in Giant Clam (Tridcana maxima).
The PO activity was measured by monitoring the absorbance at 490 nm using a microplate reader.
As in other molluscs, PO was found to exist as a proenzyme (proPO) in hemocytes.
Inhibition studies (EDTA, CPIC, PMSF and HBHZ) confirmed the proPO activating system of Giant Clam is a proteinase cascade in which Ca2+ dependent serine proteinases proteolytically convert proPO into active PO.
Activated PO was found to be a tyrosinase-like enzyme that is responsible for both monophenolase and diphenolase activity.
The bifunctional PO had the same affinity for the monophenol, hydroquinine monomethyl ether (4HA) (Km = 11.03 mM) and the diphenol, L-DOPA (Km = 11.91mM).
Maximum enzyme activity was evident at 45 °C, pH 8 and 45ppt for L-DOPA and maximum enzyme activity was evident at 30 °C, pH 9 and (30 to 40) ppt for 4HA. The results of this study were shown that phenoloxidase activity in the Giant Clam (Tridcana maxima) does provide a reliable indicator of stress.
