Three types of cell cultures, chicken embryo fibroblast (CEF), chicken kidney cells (CK) and chicken liver cells (CL) were prepared for adaptation and propagation from duck embryo adapted strain D76 of egg drop syndrome (EDS).
After five passages, the virus titre reached 109, 108 and 107 TCID50/0.1 ml in CEF, CK and CL, respectively, where the cytopathic effect of the virus began to appear at 3, 3 and 4 days post inoculation, respectively.
The studies of the virus growth curve revealed that the peak titre was detected at 72, 80 and 92 hours post inoculation in CEF, CK and CL, respectively.
Therefore, the use of cell culture system instead of embryonated duck egg system for adaptation, propagation and titration of EDS-76 is recommended because it is rapid, sensitive, specific and reduce the chance for contamination.
