English Abstract

PPOs are enzymes that catalyze the oxidation of phenolic compounds using molecular oxygen.

The ability of polyphenol oxidases to act on phenolic compounds makes them highly useful biocatalysts for various biotechnological applications.

In this study, Extraction and characterization of polyphenol oxidase from Phaseolus vulgaris L., Pisum sativum L.and Vicia faba L.

was achieved.

The enzymatic browning was determined by spectrophotometer for 100 min.

The PPO was tested by measuring the initial rate of quinone formation, as indicated by increase in absorbance at 420 nm.

Phaseolus vulgaris L.

showed the highest browning intensity when compared to the other tested plants.

Most of the browning was completed during the first hour of incubation.

The percentage of browning which occurred during the first hour of incubation was found to be 92% for Phaseolus vulgaris L., 85% for Pisum sativum L., and 80% for Vicia faba L..

Characterization studies indicated that the enzyme showed highest activity at pH 5 for Phaseolus vulgaris L., 7 for Pisum sativum L., and 6 Vicia faba L., and the optimum temperature for the PPO enzyme from Phaseolus vulgaris L., Pisum sativum L., and Vicia faba L.

was found to be 50°C, 50°C and 40°C, respectively.

The activity of PPO enzyme using catechol as substrate were found 0.917 μmol/min, 0.675 μmol/min and 1.42 μmol/min for Phaseolus vulgaris L., for Pisum sativum L., and Vicia faba L., respectively.

The Km values of the enzyme is 1.63 mM Phaseolus vulgaris L., 2.12 mM for Pisum sativum L., and 5.55 mM for Vicia faba L..
