English Abstract

A total six different bacteria were isolated from two years TNT contaminated soil samples collected from ma`an quarries , south of Jordan.

TNT is a nitroaromatic compound which have as a intermediates product 2,4-DNT and 2,6-DNT mixtures ,this mixture used in the manufacture of dyes, pesticides, polymers and explosives .

Typically manufacturing of TNT produced 76 % 2,4-DNT , 19 % 2,6 DNT ( ratio 4 : 1 ) and small amount of other compound .

This study to find and optimized bacteria that have the ability to degrade 2,4-DNT.

Many methods used to identified bacteria starting with colony morphology, gram stain, oxidase test and 16s ribosomal ( rRNA ) gene sequencing.

This bacteria cultured in BSM that have only 2,4-DNT as a carbon , nitrogen source and sole energy, were O.D.

measured at 600 nm and 418 nm every 8 hours , numerous condition studied pH, Temperature and carbon source, nitrogen source .

Two isolated bacteria show the ability to degrade 2,4-DNT , this bacteria identified by 16S rRNA as Bacillus sp.

SP79 and Bacillus lichenformis, the degradation rate was Bacillus Sp.

SP79 0.35 ( μM / h ) and Bacillus licheniformis 0.67 ( μM / h ) , growth and degradation ability show small effect by physical condition ( pH, Temperature ) and highly effect by chemical condition ( carbon and nitrogen source ) according to what bacteria prefer .
