English Abstract

Two bacteria was isolated from quarries' soil contaminated with 2,4-DNT compound for 5 years ago, from Ma'an city southern of Jordan.

It was identified by using morphological and biochemical characteristics as Acinetobacter calcoaceticus , Acinetobacter sp.

Where found to use the 2,4-DNT compound ( is an Nitroaromatic organic compound with the formula C7H6N2O4 ,which is an important intermediate in the explosive industries and considered as a U.S.

Environmental Protection Agency priority pollutant ) as alternative carbon and nitrogen source at 110 μM concentration .

degradation ability was monitored through the disappearance of the substrate and the growth of bacterial cells on the substrate.

The degradation is a catabolic process catalyzed by enzyme activity .

A dioxygenase oxidizes the nitro group either at ortho or meta position.

The degrading ability tested by using base salt media (BSM) were done in 500 ml Erlenmeyer flasks with working volume of 100 ml on a rotary shaker (37°C, 150 rpm) For 72 hour .

The best results of degradation of 2,4-DNT compound were obtained with 110μM substrate concentration ,40oC , PH 7.5 , 8 .

The carbon sources which used at ( 0.2 %) enhanced the growth and affected the biodegradation differently , glucose reduce the degradation percentage .

Nitrogen sources used enhanced the growth for 8 folds of that control ,also enhanced the biodegradation rates of 2,4-DNT at different levels .
