English Abstract
Medicinal plants are considered as a safe, complementary and alternative source for medicinally important drugs.
Aerial parts of Achilleafragrantissmawere extracted with ethyl acetate and the resulting crude extract was subjected to successive silica gel, sephadex column chromatography and purified compounds were isolated using TLC chromatography.
All cytotoxic fractions and sub-fractions arethose that were eluted with non-polar to semi-polar eluents.
A total of forty five different cytotoxic compounds purified were tested against breast adenocarcinoma cell lines; MCF-7(ER+,PR+ and HER-2+),MDA-MB-231(ER-,PR- and HER-2 -) and Vero cell lines (normal control cell).
Their effect was time and dose dependent and the anti-proliferative potential varied between IC50 of 7-49 µg/ml, 11- ˃50 µg/ml and 2- ˃50 µg/ml against MDA MB-231, MCF-7 and Vero cell lines, respectively.
Compounds 5B1,6A2,6A3, showed a selective anticancer activity against malignant cells with IC50 values range (20-30 µg/ml) with negligible effect on Vero normal cell lines at IC50˃50 µg/ml.
The Phytochemical screening indicate that purified compounds belong to flavonoid,terpenoids,steroid, sugar or phenolicgroups.
They exert their cyotoxic effect by induction of apoptotic or necrotic pathway in affected cell line.
Therefore, A.fragrantissmamay be considered as a potential source for anti-breast cancer agents with notation of the dose that may cause harmful effect on medicinal herb consumers
